Bacteriostatic Water vs. Sterile Water in the Lab

Both are clear, sterile liquids used to reconstitute peptides, but they are not interchangeable. The difference comes down to a single preservative.

When reconstituting research peptides, two solvents come up constantly: sterile water and bacteriostatic water. They look identical, and both are used to dissolve lyophilized powder. The practical difference is what allows one to be used over multiple withdrawals while the other is intended for single use.

Sterile water

Sterile water is exactly that — water that has been sterilized and contains no additives. Because it has no preservative, once a container is opened and accessed it offers no ongoing protection against microbial growth. It is generally treated as single-use.

Bacteriostatic water

Bacteriostatic water is sterile water containing a small amount of benzyl alcohol (commonly around 0.9%) as a preservative. "Bacteriostatic" means it inhibits the growth of bacteria — it does not sterilize an already-contaminated sample, but it suppresses growth, which allows a sealed multi-dose container to be accessed repeatedly over a limited period.

The benzyl alcohol is the entire difference. It is what makes bacteriostatic water suitable for stock solutions accessed more than once.

Which to use

  • For a stock solution you will draw from multiple times: bacteriostatic water is usually preferred
  • For a single preparation used immediately: sterile water is sufficient
  • For peptides sensitive to benzyl alcohol, or assays where any additive interferes: sterile water avoids the preservative

As with any reconstitution, confirm the peptide’s compatibility with your chosen solvent first, and keep reconstituted solution cold and used within a sensible window.
