Melanotan-2 (MT-2) is a synthetic cyclic analogue of alpha-melanocyte-stimulating hormone (α-MSH). It is studied because it is a broad-spectrum agonist — it activates multiple receptors in the melanocortin family rather than just one — which makes it a useful tool compound for investigating melanocortin signaling in research models.
The melanocortin receptor family
Melanotan-2 activates melanocortin receptors MC1R through MC5R. Each receptor sits on a different signaling pathway, which is why a single broad-spectrum agonist is studied across several distinct research areas:
- MC1R — central to melanogenesis (pigment) signaling, studied in melanocyte cell lines
- MC4R — associated in the literature with central appetite-regulatory pathways
- MC3R / MC5R — studied in the context of energy homeostasis and receptor pharmacology
Where it appears in research
Because of its broad receptor activity, MT-2 is used in research investigating melanogenesis signaling via MC1R, central appetite-regulatory pathways via MC4R, and energy-homeostasis receptor pharmacology. As a cyclic peptide, it is also studied for its stability relative to native α-MSH.
